Title : EFFECTIVE RAT MODEL FOR THE EVALUATION OF ADAPTOGENIC SUBSTANCES
Abstract : A modification cold-hypoxia-restraint rat model was developed for evaluating improvement in Physical work performance and endurance in adverse stressful environment, which is an essential characteristics of an adaptogen endurance to a stressful condition. A number of medicinal plants have been used in Indian system of Medicine as adaptogens, but due to lack of scientific evaluation, both experimental and clinical, these drugs have not achieved due recognition. In present investigation Asparagus racemosus (SATAVARI) root, Terminalia chebula (HARITAKI) and Emblica officinalis (AAMALAKI) fruits extracts and their fraction were evaluated for positive endurance promoting properties in this model, where restrained rats were exposed to 4 degree C at 480 mm.hg atmospheric pressure in an animal decompress chamber. In such a stressful environment, the colonic temperature of the rat eventually begins to fall. Attainment of a colonic temperature of 23oC was taken as the termination point of the exposure. At this point adrenal weight, ascorbic acid, cholesterol and brain lactic acid was estimated with time to attain 23oC and time for its recovery at room temperature. Panax ginseng (50 mg/kg, p.o. x 5 days) was used as standard for comparison. Crude extract of all the three plants studied in a dose of 200 mg/kg. p.o. for 5 days showed significant (p<0.001) adaptogenic activity (35-80%) in all the parameters. Further work is in progress to localize the active constituent in these plants.
